How to find the interaction between two proteins - (Jan/12/2015 )
Hi,
I would like to know, if there is a way to find the interactions between two proteins. Iam working on a dental protein called Dentin Matrix protein and loss of this activity is reported to increase the Fgf23 levels in vivo. But there are no reports to show how this is happening.
The problem Iam facing is when I did a promoter activity by duel luciferase assay for Fgf 23 using constructs of varying length (0.6- 2.3 kb), co transfecting cDMP1, instead of inhibiting the activity of Fgf23, it increased the activity. I dont know the reason why. This is in contrast to the reported results. Going crazy. Please help.
There are so many ways to measure protein-protein interactions. This kind of question is best answered by somebody (such as your mentor) looking at what exactly your project is and what you are trying to accomplish. Difficult to say anything short and helpful, based on your brief description. Check out these protocols and see if something works for you: http://www.protocol-online.org/prot/Molecular_Biology/Protein/Protein_Interactions/
Thanks. I wil discuss it with my boss
