co-IP stained with coomassie blue - (Jul/19/2013 )

hello everybody,

i tried to perform co-IP experiment using protein A beads and tried to stain my gel with coomassie blue just to check.

however i saw only one very strong band corresponding to approximately 55 kda. since my target protein also corresponds to around 50 Kda, i am confused whether the strong band i saw is IgG or my target protein.

there are no other bands i saw from my gel. except for that strong band at 55kDa. what could it possibly be?

That band will be heavy chain of IgG. The (co)-IP'd bands may or may not be visible, but should be detectable by specific antibody, even if they aren't visible.

you could include a no antibody control to the experiment to confirm that.
