Summary: Dicer Substrate RNAi is triggered by 27-mer dsRNA first reported by John Rossi's group, while the classic RNAi method uses 21-mer synthetic siRNA. Dicer substrate RNAi is claimed to have a higher gene knockdown efficiency compared to the classic RNAi because the 27-mer dsRNA can be processed by dicer and further incorporated by Argonaute proteins in a natural way into the RISC complex. To performed dicer substrate RNAi, synthetic 27-mer RNA duplex is used to transfect cells.
