- Protein Assay (David R. Caprette, Rice University )
A list of assays for the determination of protein concentration in a solution. This list includes the sensitivity range, volume/amount of sample needed, subjective comments on accuracy and convenience, and major interfering agents. Procedural details, equipment requirements, and references are outlined in the individual assay documents. Two categories of methods are included: Absorbance assays and colorimetric assays (Modified Lowry, Biuret, Bradford assay and Bicinchoninic Acid (Smith)).

Coomasie Blue Staining of Protein Gel

Coomasie Brilliant Blue R 250 is the most commonly used staining procedure for the detection of proteins. It is the method of choice if SDS is used in the electrophoresis of proteins, and is sensitive for a range of 0.5 to 20 micrograms of protein.

Immunolocalization of photosynthetic proteins

The protocol works well with abundant photosynthetic
cytoplasmic or chloroplast localized proteins. It should work for other abundant proteins too. For detecting rare proteins, nuclear localized proteins and possibly cell wall proteins, modifications may be needed.

Perhaps today one of the most popular non-radioactive protein detection is the silver staining which is 100-fold more sensitive than Coomassie Brilliant Blue staining

Protein Limited Proteolysis

This protocol describes the general procedure for setting up a limited proteolysis reaction. An experiment like this is useful for determining the sizes, timed order of appearance, and relative amounts of fragments produced from the protein of interest by digestion with a protease. From the results, you may be able to infer the presence of stable subdomains. It is also possible to obtain evidence of binding of one protein to another by running two identical sets of proteolysis experiments, one with the putative target, and one without.
