Southern Transfer to Nylon Membranes

Southern transfer is the classic method used to move DNA (usually in the form of restriction fragments) from agarose gels to a solid support, e. g. nylon membrane. This protocol is designed for transferring DNA for RFLP analysis. The optimum size range of DNA fragments detected this way is 2 - 25 kb.

Standard Hybridization Technique for DNA

The standard solution typically used for both pre-hybridisation and hybridisation is based on that given in Maniatis et al., (1982) with both Denhart's solution and heterologous DNA being replaced by heparin (Singh and Jones, 1984).
