Immunofluorescence Staining and Flow Cytometry of Intracellular Cytokines
(eBioscience)
A modification of the basic immunofluorescence staining and flow cytometric analysis protocol can be used for the simultaneous analysis of surface molecules and intracellular cytokines at single-cell level. In this protocol, cells are first activated in vitro, stained for surface antigens, as in the regular staining protocol, then fixed and permeabilized to allow for anti-cytokine antibodies to stain intracellularly.

Immunostaining of tissue sections
(EnCor Biotechnology)
Here are two alternative procedures, both of which work well. The more well established method is to perfuse the animal with fixative while under anesthetic. It is a lot easier to sacrifice the animal first, dissect out the tissues you want and just drop them into fixative. Fixed tissues are then impregnated with sucrose, which acts as a cryoprotectant, preventing ice crystal formation during freezing. Specimens are then rapidly frozen and sectioned on a cryostat.
