DNA Extraction from Stem Cells

detail the extraction of genomic DNA from human embryonic stem (hES) cells and induced Pluripotent Stem (iPS) cells. Cell cultures in flasks or petri dishes can be harvested as soon as sufficient numbers of cells are available (>106 cells). The cells are dissociated from the culture vessel and lysed, RNA and cellular proteins are enzymatically degraded, and the DNA is isolated from cellular components using a spin column with a silica membrane. Ultraviolet (UV) spectrophotometry and agarose gel electrophoresis are used to assess purity, concentration, and integrity of the isolated DNA.

details the G-T-L banding (G-banding) of metaphase chromosomes for cytogenetic analysis. G-T-L banding refers to the chromosome banding technique in which G-bands are acquired through treatment with Trypsin followed by staining with Leishman’s stain to produce differential staining of metaphase chromosomes. G-banding allows for pairing of homologous chromosomes and detection of structural chromosome abnormalities. G-T-L banding results in light banding patterns in euchromatic regions rich in cytosine-guanine base pairs that are generally rich in active genes. G-T-L banding results in dark banding patterns in heterochromatic regions rich in adenine-thymine base pairs that have a higher density of inactive genes.
