Cached - Cell Counting/ Live-Dead Discrimination using EB and Acridine Orange (Flow Cytometry Core Facility, University of Chicago)
This is a microscopy based application for definitive discrimination of live and dead cells. Trypan Blue exclusion notoriously over estimates the number of dead cells. Using this protocol, you can obtain a more accurate count of cells.

Cached - Counting and Determining the Viability of Cultured Cells (Journal of Visualized Experiments)
Determining the number of cells in culture is important in standardization of culture conditions and in performing accurate quantitation experiments. A hemacytometer is a thick glass slide with a central area designed as a counting chamber. Cell suspension is applied to a defined area and counted so cell density can be calculated.

Cached - Trypan Blue Viability Test (Dr. Volker Briese, Universitäts-Frauenklinik Rostock)
Trypan blue will stain dead or dying cells. Viable cells are able to repell the dye and do not stain. Note: Trypan blue has a greater affinity for serum proteins than for cellular protein. If the background is too dark, cells should be pelleted and resuspended in protein-free medium or salt solution prior to counting.

Cell Counts Using a Hemocytometer
(Donis-Keller lab)
The purpose of this procedure is to determine the cell density of the culture. Cell cultures always have some dead cells; the viable and non-viable cells can be distinguished with the use of trypan blue dye and a hemacytometer. Living cells will not take up the dye while dead cells do.
