Macrophage - Survival without changing media (Sep/30/2005 )
Hi,
for one experiment I need to keep my macrophage the more longer possible (21 days) without changing the media and with a cell death pourcentage low (like 10%). My murine macrophage are in 24 wells with RMPI.
So what do you think, what kan add or do?

according to my experience its not possible if its possible its require to add aditional extra media after some interval................
You not mention here how much media you were taking in the per well and how cell no.
we are also using some experiment where its require to mantain the macrophase in the 24- 12 well culture plates for 42-48 hrs but i seen thats they get stress if we not adding sufficent amount of media.
You not mention here by which methodes you check the % of cell death

I find my macrophages which are a by product of other cell isolations I am doing often live a very long time with little media change (like eveyr 2 weeks).

Hello thanks for your replies,
In fact I use 1 ml of RPMI per well (in 24 wells) and take 25 000 macrophage (Mf4/4, double time +/- 24h) per well also. For the beginning I just use trypan blue technic to detect the cell death duing time. I have also an other cell line called NR8383 (Rat macrophage) with a double time really long but this kind of cell is so difficult to cultivate.
