I am interested in generating stable transfections in fibroblasts and squamous epithelial cells. Can anyone suggest what protocol and which reagents to use.

The protocol you need is in a paper in J. Cell Biology 1996. 134: 985-1001. Bornslaeger et al. and it starts out as Breaking the connection: displacement of the desmosomal plaque............etc. A lot of stable cell-line guys reference this article.
