For all Western Blot users I would strongly recommend to do a Ponceau Staining of the Mebrane after the transfert to be sure that transfer was done accurately.

The good thing about ponceau staining is that it's reversible (wash away with pBS) and doesn't interfere with antibody detection !

On semi dry blotters there are from time to time some area not well transfered usually due to dryness of one side of the transfer sandwich or a big bubble between the gel and the membrane !
