Does methylation matter? - (Sep/27/2005 )

Hi everyone,

When I amplify a vector in a bacteria that is dam-methylation and dcm- methylation negative Ill get a vector that is unmethylated. smile.gif If I then try to transform another bacteria (in my case TOP10) with this unmethylated vector - will the vector be cutt to peices?
If I try to transfect eukaryotic cells with unmethylated vector will it "survive"? ph34r.gif
Thanks for sharing your knowledge.........

Should not be a problem.
