DNA PCR on blood sample - help-Genomic DNA PCR from clogged blood. (Sep/26/2005 )

I got very nice genomic DNA from partially clogged mice blood, it can be cutted by EcoR1 or BamH1, or PCR it with the house keeping gene.
For unknow reason I can not PCR out UNG gene, many UNG controls are working, is that possible the Li-heparin bind the targeted DNA?

Hi,

The Li-heparina interferes with PCR, so in my lab we extract blood for PCR in k3-EDTA and we perform PCR and qPCr without problems.

Hi,

The Li-heparina interferes with PCR, so in my lab we extract blood for PCR in k3-EDTA and we perform PCR and qPCr without problems.

thanks, since blood samples were in Li-heparing, how can i get rid of it.

clean ups wont work if the DNA is already "damaged" as Li-Heprain can do - your best bet is to re-collect your sample. Even a blood spot will work better than a Li-H sample.
