I am using perlprimer to check my methprimer designed primer sets, just want to see if there are dimers. Can somebody tell me how can I tell if those dimers shown on the programe are serious problems. I compare the results with IDT, big difference. I don't know how to justify it.

perlprimer's default temp for primer dimers is 37C if you increase this to the Tm of your PCR cycles most will disappear.

Different programs have different ways to determining Tm and dimers, so it's best to pick one and stick with it, at least you are getting consistency!
