Hi,
I need help with stoping proliferation of cells.
I am working with endothelial cells and the aim is to measure migration. However, there is a significant contribution of proliferation to migration proces.
Does anyone know how to stop proliferation without interfering with migration ability of the cells??
any other suggestion for eliminating the proliferation contribution is highly welcomed.

as i know, proliferation is highly related with growth factors present in the serum you add in your medium. It would be great to reduce the concentration of serum.
I can't tell you the right amount because i've never done it on endothelial cells. But you can reduce it at 1% level and see what hapens. 0.2%serum is used by a colleague for serum starvation assays. So it could be the limit.

Usually migration assays are done within a short window of time so proliferation isn't an issue. I suggest you shorten assay time to 4-6 hours or something.

short time assays are suitable for invasion through matrigel for example. in order to see a significant migration over a distance or in oeder to measure speed one should make kong assays for at least 24h and i saw cases where 3days was the optimal time.
one more thing how can i measure and/or visualize only proliferating cells (i am familiar with brdU assay)
