Viral RNA extraction - Formalin fixed & Paraffin embended tissue (Sep/19/2005 )

Is there any difference between viral RNA extraction kits and kits for RNA extraction sad.gif ? I f there is no difference why some companies provides those two kits ohmy.gif ??

There are some differences between RNA kit extractions. Depending what you need. The kind of sample, volume. It change Disruption/ Separation Method, etc.

FTA cards have been used to extract DNA and RNA from different samples, including virus. I heard that they work pretty well. You just put your sample on the card, let it try, take a punch and use it. For DNA, the molecule is entrapped on the card matrix, and RNA is eluted from the card.

there is the FTA elute, it's a card where you can elute DNA from the matrix.

There are some differences between RNA kit extractions. Depending what you need. The kind of sample, volume. It change Disruption/ Separation Method, etc.

I'm not sure which kit would be better for you, actually, now, I'm using trizol with good results, but I don't work with tissue samples.

If you have very small amount of sample, I think you could optimize your RT-PCR protocol too.
Some time ago, I was having problems with samples with very few copies of virus and I resolve it by changing to the one-step RT-PCR (with RT III) from invitrogene.
Now a day, how are you extracting your RNA?

Hallo, I have used ki from machaey Nagel for RNA extraction including viral RNA from lung tissue. I have nice yields and use it for real time PCR of viral mRNA. Result seem good. There is an modification of protocol for parafin fixed or else. Qiagen offer usefull kits as well

I tried several kits from different vendors for isolation viral and mRNA from my ffpe samples. In the end qiagen and roche gave best yields - however for following qRT-PCR and classical RT-PCR the Roche kit (High Pure FFPE RNA Micro Kit) worked best AND even it is cheaper than the qiagen kit smile.gif so now i always go for the highpure kit
