hi, im working on the analysis of plant metabolites, the project requires analysis of different cellular compartments for the presence of those metabolites.the block is :the tissue is frozen(-80) and is hard to get intact organeles especially vacuoles;, methods include cell fractionation but most of them are fractions and are marker dependent and full seperation is not shown.ideas from any and all are welcome.waiting .........

You could try cryosection and then laser microdissection.
