DMEM Storing - 1 month without use it (Sep/05/2005 )

Hi

I use DMEM with 10-20% FBS for my cell colture, Glutamine, pen strepto, and gentamyicin phosphate.

I left some medium at 4°C for about 1 month (400ml DMEM with 10% FBAs etc, and 200 ml only basic DMEM). I know that Glutamine must be added again because it is used to degrade with time. After this, once verified that pH is ok, Can I use this DMEM? Or must I waste it?

I've used 1 month old DMEM (stored at 4 degrees) w/ similar serum and supplements without any problems , and did not add additional glutamine. My cell line is very robust though, and yours may respond differently.

I'll happily use 1 month old DMEM (10% FBS, Pen/Strep) for my HEK293s. Works just fine for me. What cell line are you using?

In our facility, we perform cell based assays on diverse range of cell lines, all cultures are growing pretty fine.

Usually we don't use basal media which is more than three month old, and
complete media (containing serum), more than 1.5 - 2 months old.

I am working on human chondrocyte and it is a roubust cell line. I'll try to use the DMEM and if neessary I will "refill" Glutamine!

It was first time I left DMEM for so long time without use it so I found useful your suggestions smile.gif !
