I want to construct a cDNA library from coleus blumei leaves to study leaf color regulation,but I meet a question before I construct it.
I dont know how to select a appropriate time of the leaves to construct the cDNA library.

It depends on your purpose. If you would to study the enzymes that synthesizing the color compounds, you need to find out what compound it is and when/where it is made most abundantly and use that tissue type/stage for cDNA library. Becuase presumably that tissue type/stage would have more mRNA of your gene of interest, thus giving you a higher chance of cloning it.
If you are interested in the regulatory members, I don't have much idea about that. An educated guess would be making libraries of two stages of most different colors.
