design pcr primers - (Aug/24/2005 )

hello, could anyone share some experience with me about how you designed your pcr primers that contain restriction site and point mutation(s) please? I am currently struggling to obtain a design that has good melting temperature (10 degrees above the extension tm) length within 45bps and capable of avoiding dimers and hairpin.
