hello...

can we PCR DNA fragment for more than 2 kb?what is the maximum size for PCR then?

help needed..thanks.. laugh.gif

Yes we can, it all depends on the enzyme you're using. I regularly go up to 4 kb, and I have just done Site directed mutagenesis (which also has a PCR-step in there) on an 8 kb plasmid, but you can go way beyond that I believe.

WE've had luck amplifying a 11KB PCR product from genomic DNA with
LA Taq.

hi try Roche Expand, high-fidelity pcr system, im about to use it for inverse PCR of a 7kb fragment, it was recommened to me, it says that 5kb is routine in the protocol.
