what is the chemical and physical changes take place during fixing?

Heat fixing? The cells die. They decrease in size by 1/3. Lose water. The capsule, flagella, forget that you'll see them.

I guess water loss is the biggest difference. This leads to protein coagulation and similar things alongside.
