Question: Different between DMSO and Betaine - (Aug/12/2005 )

Hey everybody,
I have a question about PCR. What is the difference between DMSO and Betaine. Cause some of my DNA sample work well with Betaine but not with DMSO and vice versa. So just want to check what happen actually other than the chemistry of the two products were different. I really appreciate for those who answer my question. Thank you very much.

PCR is also a physical reaction. The viscosity of DMSO and Betaine in the PCR mixture play an important role in the thermocycling parameters. Betaine is a polymeric compound produced by plants in response to heat or saline stress so it has a physiological function in living organisms. DMSO is a synthetic compound.

I wonder the different role played by the DMSO and Betaine in PCR reaction? could you tell me ?
And which reagents is better in amplifing the GC rich sequence?
