How can I isolate RNA from fungus? How about the same protocol for plantsor the protocol for tissue? If it is possible,tell me more about the detail which is needed to notice.

I do not know about what other people use, but there are separate kits available for isolation of RNA from specifically fungi, plant, and tissue samples that our lab uses.You can try using the EZNA Fungal RNA kit from Omega Bio-Tek, Inc.that uses spin columns and is specially geared to remove mucopolysaccharides from fungi. These kits do not require the use of shredding /homogenizing accessories. The method involves a simple and rapid precipitation step for removal of most of the polysaccharides and phenolic compounds found in such tissues from as much as 200 mg tissue (in less than one hour).Plant RNA protocol is tricky depending on the source of the sample, sugar and other polysacch content and from which part of the plant the RNA needs to be extracted.Tissue RNA extraction requires a modified protocol.But I think that the EZNA Fungal RNA extraction kit should suffuce for your purposes.
