Extracting RNA from skin - Isolation and concentration (Aug/11/2005 )

Hi everyone,

I was extracting RNA from fish skin mucus and the RNA was too slow. Anyone has any better protocol to extract RNA from skin. And also to concentrate it?

Would really appreciate all your help.

hi,

i usually treat total tissu (skin, mucus, mouse tail...) in Tris 100mM, EDTA 5mMn SDS 0.2%, NaCl 200mM and proteinase K 100µg/ml
the tissu is fully digested O/N at 55C
after that i use standard TriReagent protocol beginning with 250µl of crude digested skin + 750µl TriReagent...
