RAPD PCR help - (Aug/04/2005 )
Hi,
I know this is a pretty basic question but I need to know the answer. Can you tell me if you need to use forward and reverse primer during random amplification polymorphic DNA (RAPD)?
THanks

No. But you might find that you get more reproducible results if you do - that is my experience anyway

No, RAPD normally uses only one primer, a random 10-mer which function as both forward and reverse primer. This is possible because a 10-mer sequence by chance occur relatively frequently, and thus the primer binds at random on both DNA strands.
