priciples behind nucleic acid isolation - need this for additional lecture information (Aug/02/2005 )
I really need some information on the actual principles behind the following procedures to isolate nucleic acids: alkaline lysis , phenol chloroform extrcation , cesium chloride and silica dioxide.
What actually happens? why it happens ...the chemistry details ...........

I am not going to answer all your questions, but the basis of CsCl centrifugation is differences in density between, RNA, DNA and protein. CsCl is used because it forms a spontaneous gradient when centrifuged of the right density to separate the above three fractions.
