Question about activity of restriction enzymes - (Jul/28/2005 )

Hi!

Does restriction enzymes have 5' to 3' activity, or can they read 'the other way', i.e. 3' to 5'?

don't know if my answer is the true one, but i think it's a problem of affinity. not reading in 5' to 3' or from 3' to 5', but just catch the appropriate sequence and kill it. The lovely couple of RE and target sequence...
