Alkphos Direct labeling chemiflourescent - detection with ECF (Oct/08/2001 )

Hi! I would like to know if anyone has used Amersham's 'Alkphos Direct labelling kit' before and has been successful with it. I am currently trying this kit instead of radioactivity to label my mRNA probes for my pcr blot in order to quantitate the results. If anyone has suggestions to other non-radioactive nucleic acid labelling kits that do work, I'd appreciate it if you would let me know.

There are a couple of good kits. Roche makes a good kit that uses DIG. (You then use an anti-Dig antibody conjugated to alkaline phosphatase to visualize the probes.) Roche sells whole kits that include reagents to make probes, do hybridizations, and then detect the results). Stratagene also makes a good kit that gives good incorporation results. Also, you may want to check out one of Ambion's kits. They are a company concerned almost solely with RNA. So if you want to make RNA probes, I'm sure they have some kind of labeling kit that you would find useful. Good luck!
