Maximum insert size for pUC - (Jul/08/2005 )

Hi everybody!
I cloned a 10. kb gene cluster in pUC18. Now, I have to transfer it to a shuttle plasmid (e.coli-staph), which is 5 kb long (pUC ori for E.coli). It is since 3 months that I try but it never works....
I suspect the pUC can not take up 12.5 kb? Please help me I am going mad!!!!

You would have much more luck with a low copy plasmid. Even venerable pBR322 would be better,
but a P1 or F ori based plasmid would be even better. You might consider the pSCANS inducible copy number plasmid.
