Several ways:
1. use ensembl, ensembl ( http://www.ensembl.org/Homo_sapiens/genevi...ENSG00000011304 ) genes give you all splice variant they know, shown here is the link to the PTBP1 protein, which has 3 variants;
2. check alternative splicing database ASD ( http://www.ebi.ac.uk/asd/ );
3. DIY, get all EST/cDNA of your gene, align them with your gene sequence and identify splice variants yourself.

When you say "your gene" do you mean the gDNA, unmature or mature mRNA?

Align EST to genomic sequences.

If you search for the gene isoforms of human,rat or mouse ,you can use ECgene(http://genome.ewha.ac.kr/ECgene/).
