DNA sonication !!! - (Jun/08/2005 )

I am trying to fragment salmon testes DNA (Mw = 1.3 million) using a regular ultrasonic bath.

However I red somewhere that this technique is not strong enough to break DNA chains. Is it possible ??

I'm trying to decrease DNA size (less than 500 bp) and I don't care about polydispersity because I'm just using DNA for multilayering.

So if someone knows about DNA fragmentation and can suggest me a better method to easily break DNA, I would really appreciate !!!

you could try adding some glass beads to the soup to facilitate shearing.
