genes orientation - (May/30/2005 )

hi..i wonder if the gene's orientation give effects on protein expression????

Insert orientation obviously affects protein expression. expression is possible only if the start codon sits adjacent to the promoter region present on the plaasmid. Reverse orientation will juxtaposestop codon with the pomoter. This is the reason why clones found to bear the insert ( by PCR) show negative expression.
