Transformation of an unstable gene - (May/17/2005 )

Hey all! I was wondering if anyone has some great knowledge regarding successful transformation of an unstable gene in e. coli. ...I have a gene that incurs deletions following ligation into a high copy number expression vector and subsequent transformation into e. coli. I have to use this particular vector due to the tags it possesses, and I have tried carrying out all steps of the transformation at 30 degrees. It hasn't worked yet....so I was wondering if any one had some helpful hints??? Thanks!!
