how can i culture virus from primary cell culture ?
& we are using culturing animals to culture is it desirable to cultivate & isolate viral cultures in living natural virus

As far as I understand, you want to know if you can grow wildtype virus from natural sources (primary cells/tissue) in stable cell lines and want to know if this works, yes?

Yes, this methods works when you know the virus you are incubating and have the right cells available. if you don't know what celltype your virus infects, you'll have to try some systems, till you find one that supports virus replication in vitro.
