cDNA-measuring? - (Apr/21/2005 )

How can i measure the concentration of cDNA? For measuring of RNA and plasmids i use the spectrophotometer. How about for cDNA? huh.gif

Try the spec again... cDNA is DNA afterall, so it is measureable.

a second option, try flourometer-reads only the DNA if you use Hoescht 33245. (Not to worry about the RNA in the reaction mix!)

Hello,
When you measure cDNA in your spectrophoto, you have to read absorbance at 260 and 280 as for any nucleic acid. However to calculate the concentration you've got to use a different relationchip: C (ug/ml)= 37*A260. Remember that the factor is 50 for double stranded DNA and 40 for single stranded RNA.

Is 37 for single stranded cDNA?
