DNA extraction including RNAse - (Apr/20/2005 )

we're planning a DNA extraction from blood cells via the phenol-chlorofm method. We wish to include a RNAse step, but we don't know the appropiate conditions (time, temperature, when to add...)

i don't know exactly your protocol. But usually i use to pellet dna and resspend it in TE buffer 500µl. I add 1.5µl of 20mg/ml RNase A and let do the reaction 15 to 20' at 37°C. Then add proteinase K (2.5µl of 2.5µl at 20mg/ml) and do the phenol chloroform.
