ChIP sample storage - (Mar/28/2005 )
Does anyone has the same situation?
After I sonic my sample and centrifuge them , aliquot them , put them -80c, this morning , when I dissove them on ice , they all have the precipitate.I know it bz the SDS in it, but do I need warm them to let tehm clear or centrifuge again?
I need to continue IP.

Don't worry. That's normal and it's OK to store samples after sonication.
If you let the sample warm to room temp, the precipitate should dissolve away again, although not doing this step has not affected my IP's in the past.

Hello. I am a little confused about this step. I found two ways to store the sample. One like that: after sonication, then centrifuge, divide supernatant as 200ul per aliquot, store it directly to -80 degree.
The other like that: add ChIP dilution buffer to the 200ul aliquot, then store to -80 degree. Is there any difference? or I must add ChIP dilution buffer before storing sample to -80? What do you ususally do? Thank you very much. I am naive in this field.

Well you can add dilution buffer, however it will diulte your sample, I have stored all my samples without the addition of dilution buffer. Are you using Upstates CHIP kit?

Yes, I use ChIP kit from Upstate. But unfortunately, I never made it work. There are always problems.
