MMP assay - proteases in lysis buffer (Mar/22/2005 )

I need to measure the activity of MMPs in cells. If I add protease inhibotors such as Leupeptin, apritinin and PMSF to my lysis buffer prior to lysing the cells, will these proteases inhibit the activity of MMPs?

If yes, what other protease inhibitors should I include in my buffer?

You could make cell extracts- cytoplasmic for eg, without adding protease inhibitors, as long as you work quickly and in cold conditions it shoul dbe ok.
