luciferase assay - (Mar/11/2005 )

my protocol says;

"harvest the cells by scraping for luciferase assay"

which equipment should i use to scrap my cells? won't my cells get damaged this way?

I'd say you should use a rubber policeman - don't know if that's the official term, though....

Use a cell scraper. There are different sizes dependeing on the diameter of the wells you are using. Some cells may get damaged by the scraping, but you are lysing them anyway. In my protocol for luciferase, I wash the cells with PBS and then add some microliters of lysis buffer and scrape them. So scraping may even help to lysate them.
