Inflammation measurement by PCR - How I can do it? (Feb/23/2005 )

Hello.

I want to measure inflammation on a model in which histology cant help so much... because inflammationn is very difuse.

Do you know any way to measure it by PCR?

If so, what mRNA could I use?

You can measure the level of inflammation indirectly by checking for transcription levels of certain genes by qRT-PCR. Target-cDNA is derived from mRNAs from genes for cytokines, cellular surface markers, or products of cells of the immunesystem.

usually used is a wide panel of genes like:

Interferon-alpha, -beta and -gamma
Tumor Necrosis Factor-alpha
Interleukine 1beta, -2, -4, -5, -6, -8, -12, -18, -23, -27

CD4, CD28, CD80, CD86
MHCII

iNOS

and many others. Which genes you choose is dependent on what kind of inflammation in what kind of tissue you want to study.
