How to extract DNA from halophilic bacteria? - (Jan/23/2005 )

I want to extract DNA from moderately halophilic gram negative bacteria and do PCR. Does anyone an experience in this subject? would you tell me which protocol you used?

what do you mean by "moderately halophilic"? is it found in seawater or extremly saline environment?

It was found in salty soil and needs at least 0.5 to 1 g/litter NaCl.

that's not so halophilic- try the normal protocols for bacterial DNA isolation - the classic phenol/chlorophorm extraction or commercial kits (sigma, promega, etc.) - we have used all of those for extracting DNA from marine bacteria (grown with 2 - 4 % NaCl) with no problems.
