Fc receptor binding assay - (Dec/17/2004 )

Hi
I have generated a recombinant antibody against a viral surface antigen. Now i want to check whether the recombinant human antibody can bind to human Fc receptors on PBMCs or not. The antigen that i have with me is a monomeric recombinant protein. But on virus surface it remain as multimer. Please suggest me any standard protocol to checke whether my antibody-antigen complex is able to bind to the PMBCs or not. Can i do some phagocytosis assay? If so please send me some pointers.

I'm not sure, but could a simple FACS analysis tell you if your antibody
binds non-specifically to PBMs? Your antibody should not give background higher than
the secondary antibody alone if it is not binding.
