about vectors and transfection of RNAi - (Dec/07/2004 )

primary clutured cells is very difficult to transfect or transduct in RNAi. anyone who had experience in such fields can give some suggestion about plasmid vectors and viral vectors?
1. which one is better for long RNA silence?
2.which one has a better transfection efficiency?
