Bringing tumor line from animal back to cell culture - (Dec/07/2004 )

Any thoughts? It was a spontaneously growing lymphoma that we can best ascertain. I am trying to get it to grow in cell culture from a mouse. I tried making a brei out of the tumor fragment, but it didn't grow. Now I have a actual fragment in the cell culture flask and have some growth. Usually lymphomas grow in suspension so I don't expect to have it start to adhere to the bottom of the flask. I am using DMEM and 10% FBS. Should I use another media? Perhaps some additives I am missing.

Are you trying to get a cell line from the lymphoma tissue? If so, perhaps you could incubate the tissue with cell dissocation enzymes. If you must keep media on the tissue at all times, some enzymes do work, such as collagenase (but you may need more than just collagenase).

Yes, I'm trying to isolate the cell line from a tumor fragment. What enzymes would you recommend? As of right now, the tumor fragment is laying down a base of many different cells, fibroblasts, etc. I am hoping that some of the tumor cells begin to adhere or bleb off and maintain in suspension.
