DNA Quality - (Jan/08/2009 )

I am extracting genomic DNA from buccal swabs, so getting low DNA yields (5-30ng/ul) and nothing shows up on a gel. I am using Nanodrop to check yield, and the ratios are also higher than they should be. What is the best way to check the quality of the DNA?. What is the best method to increase the concentration?. Thanks

That is REALLY low. what method do you use? I used the Qiagen kit to extract and it's good. Increase conc use a vacuum concentrator or simply leave the cap of your tube open in a clean laminar flow until the volume reduces.

Hi there,

I don't know much about buccal swabs because I have never worked with them, but could it be that weren't much cells on your swab, which led to a lower DNA yield?

Depending on how important your sample is, there is a pricy way to increase your concentration by the multiple displacement amplification (MDA) method. There are several companies offering kits using MDA for whole genome amplifications.
