Problems with CTL clones - no expansion!! :angry: (Jan/08/2009 )

Hi everybody,

I have some problems with my CTL clones. With minumal dilution I get 300-400 clones but finally only 2-10 expand. At the moment I`m using AIM-V medium with IL-2. The cells are cultured with irradiated tumor cells. I found some paper in which they used IL-7, IL-4 and/or IL-15 and sometimes they added ß-mercaptoethanol.

Does anybody have experience with this and can give me some suggestions ?

what is the detailed experimental settings for your cloning (sorting)?
are these clones from human origin?
which serum do you use? concentration of IL2?
