recently i received the proof of my article, but there is a Query(Please provide significance of “a” and “b” cited in table 1. Thank you.).

dose the editor mean i should provide the "a" and “b” when it is necessary? or i should provide the P value to the editor?

I think the reviewer asked you to do a statistical analysis (t test?) to see if there is a significant difference between a and b.

Most definately! If you have already done the statistical analysis and have an appropriate P value, why wouldn't you supply this information? The only thing it can do is support and strengthen your data.
