How to count cells on plate - (Dec/29/2008 )

Hi,

Does anyone know how to count cells on plate?

[Hi, I don´t know what exactely you want to do, but I can tell you several ways.

If you want to have a cuantitative amount of cells attached in the plate you can take a photo and then use an informatic program to count your cells.
If you want to have a cualitative result you can fix your cells and stain them with crystal violet that binds the DNA, extract it and measure its absorbance, that is proportional to the amount of cells present in your plate.

Hi,

Thank you for the answer. What I want to do is that I am culturing adherent cells. And I want to count them without detach them from the bottom.

Dear Fishman,


You need to use a Graticule on your phase contrast microscope. Different graticules have different areas. However if you count the number of cells within that area you can calculate the total number of cells on the plate/dish....but to have statistical significance count over 6 areas .......a bit of advice.....GET A COLLEGUE to do this as he will not introduce BIAS into the counting, especially if it is an experiment that YOU have set up.
