Dot blot hybridisation of RNA - (Dec/17/2008 )
Hi all....
I have labelled DIG probes(PCR product) to check the expression of three genes. Since the concentration of the probes labelled is only for 6 reactions, i planned to do dot blot of RNA. I have a problem here that is i dont have a dot or slot blot apparatus which is recommended. Which is the protocol i follow to do dot blot of RNA? Is there any protocol which says we can do dot blot without an apparatus? If there kindly share with me.

It is possible to dot blot without the apparatus, it just takes about 20 times as long and is less precise. Basically, you will need to spot small volumes (probably about 1 ul at a time) of your dilutions onto a membrane and allow to dry before spotting more on. Repeat until all your dilutions are done and then proceed as normal.
